Nursasi Handayani, M.Si.<\/p>\n<p>Kata Kunci: Fgf10, ciprofloxacin, sayap, embrio ayam (Gallus galus Domesticus)<\/p>\n<p>Ciprofloxacin merupakan antibiotik golongan fluoroquinolone yang<br \/>\nberpotensi sebagai teratogen karena memiliki berat molekul 331,4 dalton sehingga<br \/>\ndapat menembus plasenta. Ciprofloxacin dilaporkan mengakibatkan perubahan<br \/>\nstruktur tulang sumbu tubuh dan autopodium anggota gerak pada mencit.<br \/>\nPenelitian ini bertujuan untuk mengetahui efek ciprofloxacin terhadap ekspresi<br \/>\ngen Fgf10 yang berfungsi sebagai inisiator pembentukan tunas sayap, dan<br \/>\nmengatur proliferasi sel pada tunas sayap embrio ayam (Gallus gallus<br \/>\nDomesticus). Dosis ciprofloxacin 10 mg\/kg BB diberikan kepada embrio ayam<br \/>\numur 2-4 hari inkubasi. Pada umur 5 hari inkubasi, dilakukan isolasi RNA total,<\/p>\n<p>sintesis cDNA dan qPCR untuk mengetahui tingkat ekspresi gen Fgf10 dan \u03b2-<br \/>\nActin sebagai housekeeping gen. Pada umur 11 hari inkubasi dengan<\/p>\n<p>menggunakan embrio yang berbeda dilakukan pewarnaan rangka dengan Alcian<br \/>\nBlue 0,02% dan Alizarin Red 0,02%. Hasil penelitian menunjukkan bahwa<br \/>\nciprofloxacin tidak secara signifikan menurunkan ekspresi gen Fgf10 serta<br \/>\nmemicu perubahan struktur tulang pada sayap embrio ayam (Gallus gallus<br \/>\ndomesticus).<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Sari, Dwi Menita. 2017.
